Contrasto di fase a basso costo

Low cost Phase contrast

 

Una volta sfruttate le potenzialità del nostro strumento, prima o poi verrà a tutti, anche solo per curiosità, la voglia di provare almeno una delle tecniche di osservazione di alto livello..ad esempio il contrasto di fase!

Questa tecnica permette l’osservazione di oggetti o organismi incolori, e quindi invisibili con il normale campo chiaro, aumentandone considerevolmente il contrasto.

Esso normalmente si realizza mediante particolari condensatori a torretta (fig.1) che contengono all’interno una serie di aperture anulari dette “anelli di fase”i quali lasceranno passare solo un fascio di luce, appunto anulare!

 Once exploit the potentialities of our scope, sooner or later will come to everybody, the desire to try at least one of the high-level  techniques.. for instance the   phase contrast

This technique allows the observation of colorless objects or  organisms, and therefore invisible with the normal brightfield,  increasing considerably the contrast of it. 

It normally realizes  through particular turret condensers (fig.1) that  contain  inside a series of anular openings  said “  phase rings ” which will  pass only an anular light beam!

 

L’anello di luce dell’anello di fase , deve essere sovrapposto a quello di obbiettivi speciali detti appunto “obbiettivi di fase” riconoscibili per la sigla Ph (solitamente di colore verde o rossa i più vecchi) riportata all’esterno (fig.2,3)!

Da tutto ciò appare chiaro che l’equipaggiamento del contrasto di fase e tuttaltro che economico soprattutto se acquistato nuovo, per questo motivo indicherò un sistema per ottenerlo senza clamorosi esborsi!

Per gli obbiettivi di fase non credo si possa fare molto; poiché prevedono l’inserimento di un anello metallico molto  sottile di solito incollato tra le due lenti posteriori con precisione micrometrica, conviene acquistarli, magari usati su aste online e non necessariamente di marca, escludendo anche quelli ad immersione in olio…per ora;

con un po’ di fortuna si portano a casa con pochi Euro.

obvously  the equipment of the  phase contrast  is not economic especially when new,  for this motive I will point out a way  to get it without high disbursements! 

For phase objective I don’t believe can be done a lot;

 since they got a built-in thin metallic ring usually glued among the two back lenses with micrometric precision , it is worthwhile to purchase them, even used on auctions online and not necessarily of brand; 

if you are lucky you can get them for few Euro.

Le mascherine invece è possibile autocostruirsele:

Prendiamo il solito pezzo di cartoncino e il compasso e come per il campo oscuro (vedi dietro) ricaviamo un dischetto più piccolo per ottenere alla fine qualcosa come in fig.4.

Questa volta bisognerà fare molti tentativi prima di raggiungere lo scopo anche perché, come mostrato in fig.3, l’anello scuro deve contenere al suo interno l’anello luminoso, per cui è richiesta la massima precisione!

 take the usual piece of cardboard and the compass and as for the dark field (you see behind)  draw a smaller disc to get  something as in fig.4. 

This time will need to make many attempts before also reaching the purpose because, as shown in  fig.3,  the brigth ring is inside the dark one, for which it need  maximum precision.

In generale possiamo dire che esistono due tipi di contrasto di fase: il positivo e il negativo; il primo mostra il campione più scuro rispetto allo sfondo più chiaro; il secondo esattamente il contrario.

Alcune case (Zeiss Jena) producevano in passato condensatori e obbiettivi con due anelli di fase, indicati con PhV( V= variable), che permettevano il passaggio rapido dall’uno all’altro metodo..

Altri invece ( C.Baker) al posto degli anelli utilizzavano una croce che aveva il vantaggio di andar bene con tutti gli obbiettivi..

La Leitz, dal canto suo, produceva in passato il Condensatore di Heine (fortunato chi lo possiede) il quale permetteva di variare le dimensioni dell’anello di fase , semplicemente abbassando il gruppo di lenti tramite un pignone, arrivando anche al campo oscuro.

Benché io possieda un microscopio perfettamente progettato per il contrasto di fase, a volte mi diletto nel “fai da te”. Le mascherine in fig.4 funzionano ottimamente sul mio cinese che monta un condensatore di Abbe da 1,25 A.N….tuttavia mi è risultato tutto più semplice svitando la lente superiore del condensatore (fig.5) riuscendo ad ottenere un ottimo contrasto di fase anche per obbiettivi ad immersione.

Per osservare e centrare gli anelli di fase è necessario una lente di Berthrand o un telescopietto di fase che si trova sul web di tanto in tanto a prezzi modici….tuttavia non è impossibile centrare gli anelli anche senza questi ausili!

Poiché le dimensioni degli anelli di fase vanno cercate per tentativi e a volte non si sa da dove cominciare, inserisco qualche valore riferito alle misure della migliore delle mascherine che utilizzo purchè il tutto venga preso con una certa approssimazione perché non tutti i condensatori e gli obbiettivi sono uguali;

There are two types of phase contrast: positive and  negative ; the first show the sample darkened on bright background; the second (negative ph) exactly the contrary. 

Some factory (Zeiss Jena)  produced in past condensers and objective with two phase rings, marked PhV (V = variable), that  allow the rapid passage from  positive to negative phase contrast.. 

 C. Baker  instead the rings  used a cross, one for all objectives! 

 Leitz, in th past manifactured Heine condenser (you’re lucky if you got one of this)) which allowed to vary the dimensions of the phase ring, simply moving the group of lenses whit a pinion. 

Although I got a  microscope for Phase contrast capabilities, sometimes I like home made work. Phase rings in fig4 are excellent  on my chinese scope whit its   Abbe condenser whit 1,25 A.N…. nevertheless results me everything simplest unscrewing the upper lens of the condenser (fig.5) succeeding in also getting a good  phase contrast for immersion objectives. 

To centering Both fase rings ,you need of a Berthrand lens or Phase telescope, this often  came on web auctions to low price..

It’s also possibile,but more difficult, to centering phase rings whitout this!

Since the dimensions of the rings of phase must be looked for for attempts and sometimes  you don’t know from where to start, i insert some  value to the measures of the best of the phase annuli that I use, but please take it with a certain approximation because not all the condensers and the objectives are equal;

Più risoluzione dal contrasto di fase!

Come abbiamo visto il contrasto di fase aumenta la visibilità dei campioni non colorati, a volte in maniera drammatica, ma non ha alcun effetto sulla risoluzione…come migliorarla allora?

Contrasto di fase a 180°

Se invece di un anello di fase completo (fig.6) si immagina di coprirne metà o un quarto, magari sovrapponendo un’altra striscia di cartoncino e in modo da ottenere attraverso l’oculare di fase qualcosa come la fig.7,si ha un leggero ma sensibile aumento della risoluzione;

provare per credere!!

As we have seen the phase contrast   increas a lot  the contrast of the  colorless samples,  but it   have no effect on the resolution…how to improve it? 

 

180° Phase contrast

 

If instead of a complete ring of phase (fig.6) you cover half  or a quarter of it , even overlapping another strip of cardboard in order to get through the phase telescope something as the fig.7, you get sensitive increase of the resolution; 

try it to believe!!
